Overview

A combination of retifanlimab, a humanized IgG4 monoclonal antibody immune checkpoint inhibitor targeting programmed death-1 (PD-1), and INCB050465, a small molecule inhibitor of phosphoinositide 3-kinase delta (PI3Kδ), both developed by Incyte. Retifanlimab works by blocking PD-1 to release immune "brakes" and enhance T cell-mediated antitumor immune activity. INCB050465 inhibits PI3Kδ, a key kinase in B cell and immune modulation signaling, further influencing the tumor microenvironment. This combination is being investigated primarily for antineoplastic effects in advanced solid tumors, with retifanlimab now FDA approved for use with chemotherapy in metastatic squamous cell carcinoma of the anal canal (SCAC).
