This drug is a **combination regimen** consisting of retifanlimab (a monoclonal antibody targeting PD-1), pemetrexed (an antifolate chemotherapy agent), and cisplatin (a platinum-based chemotherapy). Retifanlimab acts as an immune checkpoint inhibitor by blocking the programmed cell death protein 1 (PD-1) pathway, thereby enhancing anti-tumor immune response. Pemetrexed inhibits enzymes involved in folate metabolism, leading to disruption of DNA and RNA synthesis and inducing cell death in rapidly dividing cells. Cisplatin forms DNA crosslinks, blocking DNA replication and transcription, and inducing apoptosis. The combination is being investigated to treat cancers such as non-small cell lung cancer (NSCLC) and other solid tumors, leveraging both immune checkpoint inhibition and cytotoxic chemotherapy.
