RGX-104 + ipilimumab

RGX-104 + ipilimumab is a combination therapy comprising RGX-104 (abequolixron), an orally administered small molecule agonist of the Liver X Receptor (LXR), and ipilimumab, a monoclonal antibody that inhibits Cytotoxic T-Lymphocyte Associated protein 4 (CTLA-4). RGX-104 activates expression of the APOE tumor suppressor protein, modulates immune responses, and depletes myeloid-derived suppressor cells (MDSCs), thereby enhancing anti-tumor immunity and potentially overcoming resistance to immune checkpoint inhibitors. Ipilimumab works by blocking CTLA-4, a negative regulator of T-cell activation, further boosting the immune attack against tumors. The combination is under investigation in a phase 1b/2 clinical trial for advanced cancers, including metastatic endometrial cancer and indications such as small cell lung cancer and non-small cell lung cancer[1][3].
