Drug intelligence / Profile preview
RH5.1 + AS01B (also written as RH5.1/AS01B) is an investigational asexual blood-stage malaria vaccine candidate developed by the University of Oxford. The vaccine consists of RH5.1, a recombinant soluble full-length Plasmodium falciparum reticulocyte-binding protein homolog 5 (PfRH5) antigen, formulated with GlaxoSmithKline's (GSK) liposome-based adjuvant system AS01B. The RH5.1 antigen is produced in Drosophila melanogaster S2 cells using ExpreS2ion Biotechnologies' ExpreS2 platform. PfRH5 is a highly conserved merozoite surface protein essential for the invasion of human erythrocytes via its interaction with the host receptor basigin (CD147). By eliciting neutralizing antibodies that block this interaction, the vaccine aims to prevent blood-stage P. falciparum infection and reduce parasite multiplication rates. In the Phase I/IIa VAC063 clinical trial (NCT02927145) in healthy, malaria-naïve UK adults, RH5.1 + AS01B demonstrated a favorable safety profile, robust immunogenicity, and a significant reduction in parasite growth rate following controlled human malaria infection (CHMI).
