Overview

rhIL-15 + nivolumab + ipilimumab is a combination immunotherapy regimen being evaluated by the National Cancer Institute (NCI) for the treatment of metastatic or treatment-refractory solid tumors. The regimen consists of recombinant human interleukin-15 (rhIL-15), a cytokine that stimulates the activation and expansion of CD8+ T cells and natural killer (NK) cells, combined with two immune checkpoint inhibitors: nivolumab, a monoclonal antibody targeting programmed cell death protein 1 (PD-1), and ipilimumab, a monoclonal antibody targeting cytotoxic T-lymphocyte-associated protein 4 (CTLA-4). By combining a stimulatory cytokine with the blockade of two distinct inhibitory checkpoints, the therapy aims to enhance the immune system's ability to recognize and eliminate cancer cells. Clinical evaluation in a Phase 1 trial (NCT03388632) focused on determining the maximum tolerated dose and safety profile in patients with advanced refractory cancers.
