Overview

rHIV7-shI-TAR-CCR5RZ-transduced autologous CD4+ T cells is an investigational autologous CD4-enriched T-cell gene therapy for HIV-1 infection. Patient-derived T cells are expanded ex vivo and transduced with the rHIV7-shI-TAR-CCR5RZ lentiviral vector, which encodes three anti-HIV RNA effectors: shI, a short-hairpin RNA directed against an exon shared by HIV-1 **tat/rev** transcripts; a TAR RNA decoy that sequesters HIV Tat protein; and CCR5RZ, a ribozyme designed to cleave host-cell CCR5 RNA. The combined strategy is intended to suppress HIV transcription and replication while reducing CCR5-mediated viral entry. The product was evaluated by City of Hope in a terminated pilot Phase 1 safety and feasibility study in adults with HIV-1 infection who had failed or were intolerant of highly active antiretroviral therapy.
