Ribociclib + aromatase inhibitor is a fixed combination therapy used for the adjuvant treatment of adults with hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER2)-negative stage II and III early breast cancer at high risk of recurrence. Ribociclib is a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor that blocks cell cycle progression by inhibiting CDK4 and CDK6 proteins, which are critical for cell division. Aromatase inhibitors (such as letrozole or anastrozole) block the enzyme aromatase to reduce estrogen production, thereby limiting the growth stimulus in HR-positive breast cancers. The combination has demonstrated significant improvement in invasive disease-free survival compared to endocrine therapy alone[1][2][5]. The most common side effects include neutropenia, nausea, fatigue, diarrhea, hair loss, and liver enzyme elevations[6][8][10].
