Ribociclib is an oral small molecule inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), developed as a targeted therapy for hormone receptor-positive, human epidermal growth factor receptor 2-negative (HR+/HER2-) breast cancer. It works by blocking CDK4/6 proteins, which are essential for cell cycle progression in cancer cells, thereby slowing or stopping tumor growth. Ribociclib is used in combination with endocrine therapies such as aromatase inhibitors (e.g., letrozole) or fulvestrant to treat advanced or metastatic HR+/HER2- breast cancer in adults. In premenopausal women, it may be combined with a GnRH agonist to suppress ovarian function[1][3][4][5][8]. The primary indication is advanced or metastatic HR+/HER2- breast cancer; it has also been studied for early-stage disease at high risk of recurrence[1][2].
