A combination of two targeted oral anticancer agents: **ribociclib**, a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, and **everolimus**, an inhibitor of the mammalian target of rapamycin (mTOR). This combination is being investigated for its synergistic antitumor effects in multiple tumor types where the cell cycle (CDK4/6) and PI3K/mTOR pathways are driving cancer growth, including pediatric and adult high-grade glioma (HGG), diffuse intrinsic pontine glioma (DIPG), liposarcoma, leiomyosarcoma, and other solid tumors. Both drugs are administered orally; ribociclib is typically given for 21 days of a 28-day cycle, while everolimus is administered continuously. The principal mechanism is simultaneous inhibition of cell cycle progression and tumor cell proliferation, and inhibition of mTOR signaling[4][5][3][1].
