Overview
Ribociclib + trastuzumab is an investigational combination therapy for advanced or metastatic HER2-positive breast cancer. Ribociclib is a selective cyclin-dependent kinase 4 and 6 (CDK4/6) inhibitor, while trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). The rationale for combining these agents stems from evidence that CDK4/6 pathway activation can mediate resistance to anti-HER2 therapies; thus, inhibiting CDK4/6 may resensitize tumors to HER2 blockade. Clinical studies have evaluated this combination in patients with advanced HER2-positive breast cancer who have progressed on prior standard therapies, showing manageable safety but limited efficacy in heavily pretreated populations[3][4][8].
