Ribociclib + tucatinib is an experimental oral combination therapy being studied for the treatment of HER2-positive breast cancer. Ribociclib is a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor, while tucatinib is a highly selective HER2/neu tyrosine kinase inhibitor. Both agents act via interference with cellular signaling pathways implicated in breast cancer cell proliferation and survival. The combination aims to provide an effective, chemotherapy-free treatment option by simultaneously inhibiting cell cycle progression and HER2-driven signaling in tumor cells. The combination is under investigation in early phase clinical studies, often together with trastuzumab, for use in the neoadjuvant and metastatic settings of HER2-positive breast cancer[2][3][4].
