Overview

Ridaforolimus + vorinostat is a combination therapy investigated for the treatment of advanced malignancies. Vorinostat (Zolinza) is an FDA-approved small molecule inhibitor of Class I and II histone deacetylases (HDACs), which works by increasing the acetylation of histones to promote the expression of genes that induce cell cycle arrest and apoptosis. Ridaforolimus is a small molecule inhibitor of the mammalian target of rapamycin (mTOR), a key regulator of protein synthesis, cell growth, and angiogenesis. This specific combination was evaluated in a Phase I dose-escalation study (IND 109130) led by the Fox Chase Cancer Center, targeting patients with advanced solid tumors, including renal cell carcinoma, and lymphomas that were refractory to standard treatments. The rationale for the combination is to achieve dual inhibition of critical survival pathways in cancer cells.
