Overview

This is a combination of two pharmaceutical agents, rifabutin and nevirapine. Rifabutin is an antibiotic from the rifamycin class, primarily used to prevent or treat mycobacterial infections such as Mycobacterium avium complex (MAC) in immunocompromised patients. It acts by inhibiting bacterial DNA-dependent RNA polymerase, thereby suppressing RNA synthesis and leading to cell death[2][3]. Nevirapine is a non-nucleoside reverse transcriptase inhibitor (NNRTI) used in combination antiretroviral therapy for the treatment of HIV-1 infection. It works by binding directly to reverse transcriptase, blocking RNA-dependent and DNA-dependent DNA polymerase activities including HIV-1 replication[7][10]. The combination may be considered in certain clinical scenarios where both mycobacterial infection prophylaxis/treatment and HIV management are required; however, co-administration can result in pharmacokinetic interactions—specifically, nevirapine can decrease serum concentrations of rifabutin due to induction of hepatic enzymes[1][2].
