Overview

This combination antibiotic regimen is being developed by BioVersys AG for the treatment of severe infections caused by carbapenem-resistant Acinetobacter baumannii (CRAB). The regimen consists of BV100 (an intravenous formulation of the RNA polymerase inhibitor rifabutin), the polymyxin antibiotic polymyxin B, and the siderophore cephalosporin cefiderocol. This triple-combination approach is designed to overcome multi-drug resistance mechanisms in Gram-negative bacteria. BV100 specifically targets the bacterial RNA polymerase, while polymyxin B disrupts the outer membrane and cefiderocol inhibits cell wall synthesis by utilizing the bacteria's own iron transport systems to enter the periplasmic space. The combination is currently being evaluated in Phase 2b clinical trials (RIV-CARE) for hospital-acquired bacterial pneumonia (HABP), ventilator-associated bacterial pneumonia (VABP), and bloodstream infections.
