Overview

This drug is a fixed combination regimen of three antibiotics—rifampicin, clofazimine, and dapsone—used as the World Health Organization (WHO)-recommended multidrug therapy (MDT) for the treatment of all forms of leprosy. The combination is designed to prevent the development of drug resistance and to effectively cure both paucibacillary (PB) and multibacillary (MB) leprosy. Rifampicin acts as a potent bactericidal agent by inhibiting DNA-dependent RNA polymerase in Mycobacterium leprae. Clofazimine is a phenazine dye with antimycobacterial activity; it interferes with bacterial DNA function and also has anti-inflammatory properties. Dapsone is a sulfone antibiotic that inhibits folic acid synthesis in bacteria via antagonism of para-aminobenzoic acid (PABA). This regimen has been supplied free by WHO since 1995 to endemic countries[1][2][5][8].
