Overview

This drug is a fixed-dose combination of four first-line antituberculosis agents—rifampicin (also known as rifampin), isoniazid, pyrazinamide, and ethambutol. It is used primarily for the intensive phase of tuberculosis (TB) treatment in adults and children. Each component has a distinct mechanism of action against *Mycobacterium tuberculosis*: - **Rifampicin** inhibits DNA-dependent RNA polymerase in mycobacterial cells. - **Isoniazid** inhibits synthesis of mycolic acids essential for the bacterial cell wall. - **Pyrazinamide** disrupts membrane transport and energy production in mycobacteria. - **Ethambutol** inhibits arabinosyl transferase involved in cell wall biosynthesis. The combination aims to prevent resistance development by attacking TB bacteria through multiple mechanisms. It is taken orally and included on the World Health Organization's List of Essential Medicines[1][2][4][5][6].
