This triple anti-tuberculosis drug therapy is a combination regimen consisting of rifampin, isoniazid, and ethambutol. It is currently being evaluated by the First Affiliated Hospital of Harbin Medical University for the treatment of granulomatous lobular mastitis (GLM), a benign inflammatory breast disease. The rationale for using anti-tuberculosis drugs in GLM stems from the identification of Corynebacterium species in breast tissue samples; these bacteria share structural similarities with Mycobacterium tuberculosis, such as a lipid-rich cell wall, making them susceptible to lipophilic antibiotics like rifampin. Rifampin inhibits bacterial RNA polymerase, isoniazid inhibits mycolic acid synthesis, and ethambutol disrupts cell wall assembly by inhibiting arabinosyltransferase. The regimen is being studied as a standalone pharmacological intervention and in combination with surgical excision.
