Overview

RIGVIR + nivolumab is a combination therapy being developed by Rigvir Holding that pairs RIGVIR (an oncolytic ECHO-7 virus) with nivolumab (a PD-1 monoclonal antibody). RIGVIR is a non-genetically modified, live, oncotropic virus from the Picornaviridae family that selectively infects and destroys tumor cells, a process known as oncolytic virotherapy. Nivolumab is a humanized IgG4 monoclonal antibody that blocks the Programmed Death-1 (PD-1) immune checkpoint, thereby enhancing the immune system's ability to recognize and attack cancer cells. The synergy between the two agents is intended to use the virus-induced immunogenic cell death to prime the tumor microenvironment for improved response to checkpoint blockade. This combination is primarily being explored for the treatment of skin melanoma, particularly Stage IIC, where case reports have demonstrated clinical responses including the disappearance of liver lesions and reduction in lymph node size.
