Overview

**Rilotumumab + mitoxantrone + prednisone** is an investigational combination regimen evaluated for the treatment of castration-resistant prostate cancer, especially in patients who are refractory to taxane-based chemotherapy. **Rilotumumab** is a fully human monoclonal antibody targeting hepatocyte growth factor (HGF), thereby inhibiting the HGF/c-MET pathway, which is involved in tumor cell proliferation and survival. **Mitoxantrone** is a cytotoxic anthracenedione antineoplastic agent that intercalates into DNA and inhibits topoisomerase II, leading to DNA damage and cell death. **Prednisone** is a synthetic corticosteroid that is used for its anti-inflammatory and immunosuppressive properties and as a palliative agent in prostate cancer to reduce symptoms and tumor-related complications. This triplet combination was studied in multicenter, randomized phase II trials. Although well tolerated, it did not show a significant improvement in overall survival or progression-free survival versus mitoxantrone and prednisone alone[1][2][3][4].
