This drug is a combination of three antiretroviral agents used in the treatment of HIV-1 infection. Rilpivirine is a second-generation non-nucleoside reverse transcriptase inhibitor (NNRTI) that inhibits HIV-1 reverse transcriptase, thereby blocking viral replication[7]. Cobicistat acts as a pharmacokinetic enhancer by inhibiting cytochrome P450 3A (CYP3A) enzymes, thereby increasing the plasma concentrations and effectiveness of darunavir without having direct anti-HIV activity itself[6][10]. This combination has been studied as an effective two-drug regimen for maintenance therapy in people with suppressed HIV viral load and offers an alternative to regimens containing nucleoside reverse transcriptase inhibitors (NRTIs), potentially reducing NRTI-associated toxicities[1][2][7].
