Drug intelligence / Profile preview
Rilvegostomig + FLOT is a combination therapy under clinical investigation for gastroesophageal cancers[6][2][4][7]. It consists of **rilvegostomig**, a bispecific monoclonal antibody targeting both PD-1 (Programmed cell death protein 1) and TIGIT (T cell immunoglobulin and ITIM domains)[8], and the **FLOT chemotherapy regimen**, which includes 5-fluorouracil (5-FU), leucovorin, oxaliplatin, and docetaxel[6]. Rilvegostomig blocks immune checkpoints PD-1 and TIGIT, releasing the inhibition of T cell anti-tumor activity. The FLOT regimen is cytotoxic chemotherapy targeting rapidly dividing cells and is an established standard for perioperative treatment of locally advanced and resectable gastroesophageal adenocarcinoma[6][5]. The combination is being evaluated as neoadjuvant (pre-surgery) and adjuvant (post-surgery) therapy in clinical trials for gastric/gastroesophageal cancers[2][6][4][7].
