Overview

**Rimonabant + nicotine** refers to the combined use of two active pharmaceutical ingredients: rimonabant, a selective cannabinoid receptor type 1 (CB1) inverse agonist, and nicotine, an agonist at nicotinic acetylcholine receptors. Rimonabant was developed as an anorectic anti-obesity drug that also demonstrated efficacy in aiding smoking cessation by modulating the endocannabinoid system, which is implicated in the reward pathway for nicotine. Nicotine replacement therapies (such as patch, gum, lozenge, or inhalation) are approved pharmacological agents for smoking cessation that act on nicotine receptors to reduce withdrawal symptoms. Although there is preclinical and clinical evidence for rimonabant's utility in smoking cessation, and both agents have been studied for their effects on tobacco dependence, the combination is not known to be available as a single marketed formulation. Any use of rimonabant with nicotine would be considered off-label or investigational. Rimonabant was withdrawn from the market due to psychiatric side effects including depression and suicidality[1][2][4].
