rindopepimut + sargramostim

rindopepimut + sargramostim is a combination immunotherapy consisting of rindopepimut, a peptide vaccine targeting the epidermal growth factor receptor variant III (**EGFRvIII**), and sargramostim (**GM-CSF**, granulocyte-macrophage colony-stimulating factor), which acts as an adjuvant. Rindopepimut is a 14-mer peptide spanning the EGFRvIII mutation site conjugated to keyhole limpet hemocyanin, and is designed to elicit an immune response specifically against EGFRvIII-expressing tumor cells. Sargramostim enhances antigen presentation and immune activation by stimulating dendritic cells and monocytes. The combination is primarily developed for patients with EGFRvIII-positive glioblastoma multiforme (**GBM**), aiming to improve progression-free and overall survival by eliminating EGFRvIII-expressing cells, increasing EGFRvIII-specific antibody titers, and generating humoral and cellular immune responses. The combination is administered by intradermal injection, with clinical studies demonstrating immunogenicity, improved survival versus controls, and acceptable safety.
