rintatolimod + aspirin + interferon alfa-2b is a combination immunotherapy regimen developed by Roswell Park Comprehensive Cancer Center for the neoadjuvant treatment of localized prostate cancer. The regimen, often referred to as a chemokine-modulating (CKM) therapy, consists of rintatolimod (a synthetic double-stranded RNA and TLR3 agonist), enteric-coated aspirin (a COX inhibitor), and recombinant interferon alfa-2b. This combination is designed to reprogram the tumor microenvironment by increasing the production of T-cell-attracting chemokines and decreasing immunosuppressive factors. By mimicking a viral infection through TLR3 activation and inhibiting prostaglandin synthesis, the regimen aims to enhance the infiltration of cytotoxic T lymphocytes into the tumor, potentially improving surgical outcomes and reducing the risk of recurrence in patients undergoing radical prostatectomy.
