Overview

Rintatolimod + pembrolizumab is an investigational combination immunotherapy regimen being evaluated for the treatment of refractory, metastatic, microsatellite stable (MSS) colorectal cancer. Rintatolimod (Ampligen) is a synthetic double-stranded RNA (dsRNA) molecule that acts as a selective Toll-like receptor 3 (TLR3) agonist. It functions as a chemokine-modulating agent, designed to 'inflame' the tumor microenvironment by inducing the production of specific chemokines that attract cytotoxic T lymphocytes (CTLs) and increasing the expression of PD-L1. Pembrolizumab (Keytruda) is a humanized monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor on T cells, blocking its interaction with PD-L1 and PD-L2 to restore anti-tumor immune responses. The combination is being studied by Roswell Park Comprehensive Cancer Center to overcome the resistance of MSS colorectal tumors to standard checkpoint inhibitor monotherapy by converting 'cold' tumors into 'hot' tumors.
