RIV-8 is a gene reprogramming therapy candidate developed by MedGene Therapeutics, specifically designed to address the challenge of T-cell exhaustion in cancer immunotherapy. The technology utilizes gene reprogramming to prevent the functional decline of T cells and reinvigorate those that have already entered an exhausted state. This approach is intended to enhance the efficacy of various adoptive cell therapies, including CAR-T, TCR-T, and tumor-infiltrating lymphocyte (TIL) approaches, as well as to complement checkpoint inhibitor treatments. RIV-8 employs a versatile delivery platform that includes adeno-associated virus (AAV) vectors, mRNA-based delivery, and nanoparticle formulations to modulate genetic pathways associated with T-cell dysfunction.
