RO5045377 + cytarabine is an investigational combination therapy for acute myeloid leukemia (AML) in which RO5045377, a small-molecule inhibitor developed by Roche, is administered together with cytarabine, a well-established antimetabolite chemotherapy. RO5045377 is an oral agent, with phase 1 studies evaluating its maximum tolerated dose, safety, and efficacy in combination with cytarabine in AML patients. Cytarabine acts primarily by inhibiting DNA polymerase, leading to the cessation of DNA synthesis and ultimately inducing apoptosis in rapidly dividing cells. The combination is being studied due to the potential for synergistic effects on leukemic cell proliferation and improved anti-leukemic activity.
