Overview

RO5083945 + cisplatin + gemcitabine is an investigational combination regimen in oncology trials, primarily developed for the treatment of advanced or recurrent non-small cell lung cancer (NSCLC) of squamous histology. RO5083945 (also known as GA201) is a glycoengineered, human monoclonal antibody targeting the Epidermal Growth Factor Receptor (EGFR). It antagonizes EGFR by binding and blocking ligand-induced activation, and it is designed to enhance antibody-dependent cellular cytotoxicity (ADCC) via increased binding to FcγRIIIa on immune effector cells, improving tumor cell killing compared to other anti-EGFR mAbs like cetuximab. Cisplatin is a platinum-based chemotherapeutic agent that kills cancer cells by cross-linking DNA, and gemcitabine is a nucleoside analog chemotherapy that inhibits DNA synthesis[3][4].
