Overview

RO5323441 (also known as TB403) is a humanized recombinant immunoglobulin G isotype 1 (IgG1) monoclonal antibody that specifically targets placenta growth factor (PlGF), a member of the vascular endothelial growth factor (VEGF) family involved in tumor angiogenesis. It does not cross-react with VEGF itself. Bevacizumab is a well-established humanized monoclonal antibody targeting vascular endothelial growth factor A (VEGF-A), inhibiting angiogenesis and tumor vascularization. The combination of RO5323441 and bevacizumab has been investigated primarily in patients with recurrent glioblastoma, aiming to simultaneously inhibit both PlGF and VEGF-A pathways to enhance anti-tumor efficacy by more comprehensively blocking proangiogenic signaling. Clinical studies have shown that this combination was generally well tolerated, with pharmacokinetic interactions observed—specifically, co-administration decreased the clearance of RO5323441.
