RO5429083 + cytarabine is an investigational combination therapy for hematologic malignancies, notably acute myeloid leukemia. RO5429083 (also known as RG7356) is a humanized IgG1 monoclonal antibody targeting CD44, a cell surface glycoprotein implicated in tumor growth, metastasis, and resistance pathways. Cytarabine is a well-established nucleoside metabolic inhibitor (antimetabolite) that inhibits DNA synthesis, primarily used in acute myeloid leukemia. This combination has been evaluated in phase 1 clinical trials, with preclinical data supporting a synergistic anti-leukemic effect through targeting of both proliferative and survival pathways in leukemia cells.
