RO6927005 + gemcitabine + nab-paclitaxel is a combination therapy studied in Phase 1 for patients with mesothelin-positive metastatic and/or locally advanced malignant solid tumors, especially pancreatic ductal adenocarcinoma and mesothelioma. RO6927005 (previously known as LMB-100) is a recombinant immunotoxin targeting mesothelin, a cell-surface protein overexpressed in several cancers. Gemcitabine is a nucleoside analog used as an antimetabolite chemotherapeutic. Nab-paclitaxel is nanoparticle albumin-bound paclitaxel, a microtubule inhibitor chemotherapeutic[2][4]. The combination leverages targeted immunotoxin-mediated cytotoxicity (mesothelin), DNA synthesis inhibition (gemcitabine), and anti-mitotic activity (nab-paclitaxel) to enhance anti-tumor effects.
