RO7300490 + atezolizumab is an investigational combination therapy for advanced and/or metastatic solid tumors. RO7300490 is a novel protein therapeutic with two functional domains. The first domain binds **fibroblast activation protein-α (FAP)**, commonly expressed in solid tumors, facilitating tumor localization. The second domain is a **CD40 agonist**, which stimulates immune cells within the tumor microenvironment to induce killing of tumor cells[1][2][6]. Atezolizumab (Tecentriq) is a humanized monoclonal antibody that binds **programmed death-ligand 1 (PD-L1)**, inhibiting its interaction with PD-1 and thus preventing tumors from suppressing immune responses[1][2][7]. The rationale for the combination is that RO7300490 activates immune cells to kill tumor cells, while atezolizumab blocks PD-L1-mediated immune resistance, together enhancing anti-tumor immunity[1][2]. This combination has been studied in Phase 1 trials (NCT04857138, WP42627) in patients with solid tumors unresponsive to standard therapies[1][2][3][4]. RO7300490 is an experimental agent and not yet approved by regulatory authorities for any indication. This trial is the first-in-human investigation for RO7300490 in combination with atezolizumab[1][2].
