RO7445482, also known as xalnesiran or RG6346, is a liver-targeted, GalNAc-conjugated short-interfering RNA (siRNA) designed to inhibit hepatitis B virus (HBV) gene expression by targeting the S open reading frame of the HBV genome. When combined with a nucleos(t)ide analogue (e.g., entecavir, tenofovir disoproxil fumarate, or tenofovir alafenamide), it is under investigation as a combination therapy for chronic hepatitis B infection. The nucleos(t)ide component acts as a direct-acting antiviral that inhibits HBV reverse transcriptase, thus suppressing viral replication, while RO7445482 reduces HBV gene expression to lower viral antigen production. This combination aims to achieve a functional cure in chronic HBV-infected patients.
