Overview

This is an **investigational four-drug combination therapy** comprising:\n- RO7496353 (a monoclonal antibody targeting latent transforming growth factor-beta 1, TGF-β1, being studied to modulate the tumor microenvironment and enhance anti-tumor immunity)\n- atezolizumab (a monoclonal antibody checkpoint inhibitor of PD-L1, used for immuno-oncology)\n- nab-paclitaxel (nanoparticle albumin-bound paclitaxel, a microtubule inhibitor approved for various solid tumors, including pancreatic cancer)\n- gemcitabine (a nucleoside analog antimetabolite chemotherapy)\nThis combination is designed to *synergistically* inhibit tumor immune evasion and proliferation in advanced solid tumors. RO7496353 specifically blocks activation of latent TGF-β1, thereby limiting immunosuppression and enhancing the efficacy of immunotherapy and cytotoxic chemotherapy. The combination is in early-stage clinical development (Phase 1b) for locally advanced or metastatic solid tumors such as pancreatic ductal adenocarcinoma, non-small cell lung cancer, and urothelial carcinoma.[1][3][5]
