RO7795081 + cyclosporine is a combination therapy investigated by Roche for the treatment of moderate-to-severe plaque psoriasis. RO7795081 is a small molecule inhibitor of Tyrosine Kinase 2 (TYK2), a member of the Janus kinase (JAK) family that mediates signaling for cytokines such as IL-23, IL-12, and Type I interferons, which are critical in the pathogenesis of psoriasis. Cyclosporine is a well-established calcineurin inhibitor that suppresses T-cell activation by inhibiting the transcription of IL-2 and other cytokines. The combination was evaluated in Phase 1 clinical trials (e.g., NCT03115359) to assess the safety, tolerability, and pharmacokinetics of RO7795081 when administered alongside cyclosporine, particularly to understand potential drug-drug interactions and additive immunosuppressive effects. Development of RO7795081 appears to have been discontinued by Roche.
