Overview

Robatumumab + carboplatin + paclitaxel is a **combination regimen** investigated for its antineoplastic effects in certain cancers. Robatumumab is a fully human monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), blocking the binding of IGF-1 and IGF-2, inhibiting receptor autophosphorylation, and subsequently reducing downstream signaling required for tumor cell proliferation and survival. Carboplatin is a platinum-based chemotherapeutic agent that forms DNA adducts, leading to apoptosis. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division. This combination was evaluated in clinical trials, particularly for advanced solid tumors, sarcomas, and colorectal cancer. The combination did not achieve regulatory approval, and development has been discontinued after early phase trials, with robatumumab's oncology program terminated due to limited clinical efficacy and sponsor reprioritization[1][2][3][4][5][6][7].
