Overview

Robatumumab + gemcitabine is a combination therapy comprising robatumumab, a fully human monoclonal antibody that targets and inhibits insulin-like growth factor 1 receptor (IGF-1R), and gemcitabine, a nucleoside analog and antimetabolite chemotherapy agent. Robatumumab is designed to block IGF-1R signaling implicated in tumor cell proliferation and survival, particularly in sarcomas, while gemcitabine inhibits DNA synthesis in rapidly dividing tumor cells. The rationale behind the combination is the potential for enhanced anti-tumor activity via complementary mechanisms—immune-mediated and cytotoxic action. This regimen may be under investigation or considered for solid tumors and refractory sarcoma, but is not an established approved combination as of the current evidence[1][3][7].
