This is a **combination regimen** comprising **rocatinlimab**, **topical corticosteroids (TCS)**, and **topical calcineurin inhibitors (TCI)**, being clinically studied for the treatment of **moderate-to-severe atopic dermatitis** (AD). **Rocatinlimab** is a fully human, non-fucosylated IgG1 monoclonal antibody that targets and blocks the **OX40 receptor (CD134)** expressed on activated effector and memory T cells, reducing T-cell driven inflammation and preventing the persistence of memory T cells that perpetuate AD. TCS (such as hydrocortisone or betamethasone) and TCI (such as tacrolimus or pimecrolimus) are guideline-standard, locally-acting immunomodulators used to suppress skin inflammation in AD. Clinical trials (ROCKET program) are evaluating the combination for efficacy, long-term disease control, and safety, particularly for patients inadequately controlled by topical therapies alone or for those with severe, recalcitrant disease.
