Overview

Ronacaleret + plerixafor is an investigational combination therapy that was evaluated for hematopoietic stem cell (HSC) mobilization. Ronacaleret (SB-751689) is a small molecule calcium-sensing receptor (CaSR) antagonist developed by GSK, while plerixafor is an approved small molecule CXCR4 antagonist. The combination was studied in a Phase I clinical trial (Study CR9116610) to determine if ronacaleret's ability to stimulate parathyroid hormone (PTH) release and activate bone metabolism could synergistically enhance the mobilization of CD34+ hematopoietic stem cells into peripheral blood when used alongside plerixafor. In the study, healthy volunteers received oral ronacaleret for five days followed by a single subcutaneous dose of plerixafor. However, the trial demonstrated no statistical difference in CD34+ cell mobilization compared to placebo plus plerixafor, leading to the discontinuation of this combination's development for stem cell mobilization.
