Overview

Ropivacaine + poloxamer 407 is a sustained-release, thermosensitive hydrogel formulation of the local anesthetic ropivacaine. It utilizes poloxamer 407, a biocompatible triblock copolymer that exhibits reverse thermal gelation, remaining a liquid at room temperature and transitioning into a semi-solid gel at physiological temperatures. This characteristic allows the formulation to be injected or applied as a liquid into surgical sites, where it forms a drug reservoir that slowly releases ropivacaine over an extended period (typically up to 72 hours). The primary indication is the management of postoperative pain, aiming to provide long-lasting local analgesia and reduce the requirement for systemic opioids. The formulation was notably investigated in clinical trials by Innocoll for applications such as hemorrhoidectomy, though development has primarily focused on the related collagen-based sponge formulation, Xaracoll.
