RP001 (Reach Pharmaceuticals)

RP001 is an orally bioavailable small molecule inhibitor of the Transforming Growth Factor-beta receptor I (TβRI), also known as Activin receptor-like kinase 5 (ALK5). Developed by Reach Pharmaceuticals, RP001 is designed to selectively block the TGF-β signaling pathway, which is frequently exploited by tumors to drive immune evasion, promote epithelial-mesenchymal transition (EMT), and induce fibrotic changes in the tumor microenvironment. By inhibiting TβRI kinase activity, RP001 aims to reverse these immunosuppressive effects and enhance the activity of anti-tumor immune cells, such as T cells and NK cells. The drug is currently being evaluated in early-phase clinical trials for the treatment of advanced or metastatic solid tumors, both as a monotherapy and potentially in combination with other oncology agents.
