**RP2 + nivolumab** is an investigational combination therapy of a genetically modified oncolytic herpes simplex virus type 1 (HSV-1) known as RP2 and the immune checkpoint inhibitor nivolumab (a PD-1 blocking antibody). RP2 is engineered to directly destroy tumor cells and express an anti-CTLA-4 antibody within the tumor microenvironment to stimulate anti-tumor immune responses. Nivolumab blocks PD-1 to enhance T-cell activity against cancer. The combination aims to synergistically increase anti-tumor immunity, particularly in advanced, metastatic, and immune checkpoint inhibitor-naïve solid tumors, including metastatic uveal melanoma. RP2 is administered via intratumoral injection, and nivolumab via intravenous infusion. The combination is being studied in phase 1 and phase 2/3 clinical trials for various solid tumors, especially metastatic uveal melanoma[1][2][3][4][6][8].
