RSO-021 + paclitaxel is an investigational **combination therapy** for the treatment of malignant pleural effusion (MPE) in patients with advanced solid tumors, including mesothelioma, breast, ovarian, and non-small cell lung cancers. RSO-021 is a novel small molecule inhibitor of mitochondrial peroxiredoxin 3 (PRX3) that acts by covalently inactivating PRX3, leading to increased oxidative stress and selective tumor cell death. Paclitaxel is a widely used chemotherapeutic that stabilizes microtubules, thereby inhibiting cell division and inducing apoptosis. In clinical trials, RSO-021 is administered weekly via intrapleural infusion, while paclitaxel is administered systemically, in accordance with standard of care. The combination aims to enhance anti-tumor effects via complementary mechanisms: RSO-021 disrupts cellular redox balance specifically within mitochondria of cancer cells, while paclitaxel disrupts cell division[1][2][3][5][7][9]. The combination is currently being evaluated in phase 2 clinical studies for efficacy and safety in patients with MPE due to advanced metastatic solid tumors.
