RT001 is an investigational topical gel formulation of botulinum toxin type A developed by Revance Therapeutics. It was designed as a non-injectable alternative for aesthetic and therapeutic applications, utilizing a proprietary TransMTS® peptide delivery system to facilitate the transcutaneous transport of the botulinum toxin molecule across the skin barrier. The drug's mechanism of action involves the inhibition of acetylcholine release at the neuromuscular junction by cleaving the SNAP-25 protein, thereby inducing localized muscle relaxation. RT001 was primarily evaluated for the treatment of moderate to severe crow's feet lines (lateral canthal lines) and was also studied for hyperhidrosis and migraine. However, in June 2016, Revance announced that RT001 did not meet its primary or secondary endpoints in the Phase 3 REALISE trial for crow's feet, leading to the discontinuation of the program as the company shifted focus to its injectable candidate, RT002 (daxibotulinumtoxinA for injection).
