RTX-240 + pembrolizumab is an experimental combination therapy designed for the treatment of advanced solid tumors, particularly in patients who are relapsed or refractory to prior anti-PD-1 or PD-L1 therapies. **RTX-240** is a genetically engineered, enucleated red blood cell expressing both the **4-1BB ligand (4-1BBL)** and an **IL-15/IL-15Rα fusion protein (IL-15TP)** on its surface, aiming to activate and expand natural killer (NK) cells and T cells for a broad anti-tumor immune response. **Pembrolizumab** is a humanized monoclonal IgG4 antibody targeting **programmed cell death protein 1 (PD-1)**, blocking its interaction with PD-L1 and PD-L2 to prevent immune evasion by tumor cells. The combination is under clinical investigation to assess safety, tolerability, pharmacodynamics, and anti-tumor activity, based on the hypothesis that stimulating innate and adaptive immunity via co-stimulation and checkpoint inhibition may overcome resistance to immunotherapy[1][2][3][4].
