The ruthenium(II)-thymine complex, specifically identified by the formula [Ru(PPh3)2(Thy)(bipy)]PF6, is a small molecule metallodrug currently in preclinical development for its potent anticancer properties. It has demonstrated significant cytotoxic activity against various malignancies, including acute myeloid leukemia (AML) and colon carcinoma (HCT116). The compound's mechanism of action involves direct binding to DNA, leading to DNA damage and double-strand breaks. This damage triggers caspase-mediated apoptosis through the activation of the JNK, p38, and ERK1/2 MAPK signaling pathways, often in a p53-independent manner. Additionally, the complex has been shown to inhibit NF-κB signaling and suppress leukemia stem cells, effectively reducing tumor mass in xenograft models.
