Overview

ruthenium(quinoline)2 (Ru(quin)2), chemically known as bis(quinolin-8-olato)bis(triphenylphosphine)ruthenium(II), is a novel ruthenium(II)-based coordination complex incorporating an 8-hydroxyquinoline moiety. It is being investigated as a potential chemotherapeutic agent for the treatment of breast cancer, including estrogen receptor-positive (ER+) and triple-negative breast cancer (TNBC). Ru(quin)2 exhibits antiproliferative and cytotoxic activity by inducing apoptosis, stimulating autophagy, and causing cell cycle arrest. Its apoptotic mechanism is associated with the upregulation of BAX, activation of caspase-3, downregulation of Aurora B kinase (AURKB), and histone release. Additionally, it induces autophagy (evidenced by LC3-I to LC3-II conversion and SQSTM1 reduction) and triggers G0/G1 or G2/M cell cycle arrest.
