RVU120 + venetoclax is a combination therapy under clinical investigation for the treatment of relapsed or refractory acute myeloid leukemia (AML). RVU120 is an orally bioavailable small molecule inhibitor of cyclin-dependent kinases 8 and 19 (CDK8/19), developed by Ryvu Therapeutics. Venetoclax is a well-established BCL-2 inhibitor (marketed as Venclexta by AbbVie). The combination leverages RVU120’s ability to repress AML oncogenic pathways and overcome resistance to venetoclax by downregulating anti-apoptotic protein MCL-1. This combination has shown synergistic anti-leukemic effects in preclinical models and promising preliminary activity and tolerability in early-phase clinical trials for patients with relapsed/refractory AML, particularly in those who have failed prior venetoclax and hypomethylating agent therapy. The combination is undergoing dose-finding and efficacy evaluation in ongoing phase 2 clinical studies[1][2][3][5].
