RX-0201 + everolimus is an investigational combination therapy consisting of RX-0201 (also known as Archexin), a 20-mer antisense oligonucleotide that binds AKT-1 mRNA, thereby inhibiting AKT-1 protein expression, and everolimus, an oral inhibitor of the mammalian target of rapamycin complex 1 (mTORC1). RX-0201 downregulates the PI3K/AKT pathway, and everolimus inhibits mTOR, a downstream effector in this pathway. The combination is designed to target the AKT-1/mTOR pathway synergistically to inhibit cancer cell proliferation and survival. This combination has primarily been evaluated in metastatic renal cell carcinoma (mRCC) in patients who previously progressed on VEGFR inhibitors, with early clinical studies demonstrating tolerable safety and promising clinical activity[1][5][3]. RX-0201 was developed by Rexahn Pharmaceuticals.
