S-1 + paclitaxel + cisplatin is a combination chemotherapeutic regimen comprising three agents: S-1, paclitaxel, and cisplatin. S-1 is an oral fluoropyrimidine derivative that acts as an antimetabolite inhibiting thymidylate synthase, leading to impaired DNA synthesis. Paclitaxel is a microtubule-stabilizing agent that disrupts mitosis by promoting tubulin polymerization and blocking cell division. Cisplatin is a platinum-based alkylating agent that forms DNA adducts, inducing apoptosis by causing DNA cross-linking and damage. This regimen is primarily utilized for advanced gastric cancer and has demonstrated efficacy and tolerability in Phase II clinical trials as a first-line treatment for metastatic or recurrent disease. Key reported adverse effects include neutropenia, nausea, and diarrhea. The regimen is intended for use in oncology settings and is generally administered as part of systemic therapy for solid tumors[3].
